The review by Rajman, Chwalek & Sinclair (Cell Metabolism, 2018) gathers much of the in-vivo evidence for why NAD+ matters and what happens when it is restored in animal models.

Can you raise NAD+ again?

Because NAD+ itself is poorly absorbed by mouth, the practical approach is to supply a building block the body can convert. In a randomised, placebo-controlled study in Frontiers in Aging (2022), 300 mg of NMN daily increased the blood NAD+/NADH ratio by roughly 38% from baseline over 60 days, well above placebo (Frontiers in Aging, 2022).
